Placing Your Order
Follow our three easy steps to preparing and placing your order with us.
1. Prepare your order
Before you submit your order, please make sure you prepare it correctly. Our guides are designed to help you prepare each part of your order.
Preparing your template
One of the most important factors affecting the results obtained is the quality of the DNA template. Please prepare your template using our checklist:
- Be sure that the kit you are using specifies automated DNA sequencing is a downstream application for DNA processed in that manner.
- Submit your DNA templates in either sterile water or 10mM Tris pH 8.0.
- Do not submit DNA in TE or containing gel tracking dye.

Need purification kits?
Many kits are available for purification of DNA, both plasmid DNA and DNA fragments.
We've prepared a list of recommended purification kits you may wish to select from:

Selecting primers
Please review our available stock primers, and indicate on your order if you would like to use one or more stock primers.
When preparing primers, please use our primers checklist:
- Please be sure to supply primers in either 10mM Tris-HCl pH 8.0 or in water.
- Primers should have a melting temperature of at least 50oC to work well in the cycle sequencing reactions.
- Primers should be suppled at a concentration of 5uM.

Available primers
The UMaine DNA Sequencing Facility supplies 22 stock primers at no additional charge.

Measuring concentrations
Please review our concentration measurements and follow our checklist for measuring your concentrations:
- DNAs should be supplied in either 10 mM Tris-HCl pH 8.0 or in water but not in TE.
- Concentration should be at least 100 ng/ul and 1ug will be used per reaction if attempting to sequence very large double-stranded template DNAs.

Sending your package
Before you send your package, please review our shipping checklist:
- All DNAs, template or primer, should be supplied in 10 mM Tris-HCl, pH 8.0 or in water-- not in TE.
- All tubes should be clearly labeled, sealed securely and must be accompanied by a completed order form available at the next step.
